Assuming you've exported your settings to Inner Space and your Key Maps are enabled (SHIFT+ALT+M), when you press ALT+G ISBoxer is sending G to all of your clients. If G is not set as the key binding in World of Warcraft to perform Interact with Target (in each and every game client) then nothing will happen.

That's because the default in-game key binding for your talent pane is N. You seem to not be understanding that you need to assign an Interact with Target key binding in World of Warcraft because this action is not bound to anything by default in the game itself. Here are the instructions you should have first followed direct from the Wiki page (this is also shown in the video you watched):

Finding Interact with Target in the in-game Key Bindings window
    1. Enter the game
    2. Turn Key Maps off (Shift+Alt+M)
    3. Turn Repeater on (Shift+Alt+R)
    4. Open up the in-game menu with Video, Interface, Key Bindings and other options (Escape)
    5. Click on Key Bindings
    6. Locate the Interact with Target key binding, under Targeting
    7. Set the Interact with Target key binding to your desired key combination, such as 'G'
    8. Turn Repeater back off (Shift+Alt+R)
    9. Turn Key Maps back on (Shift+Alt+M)
Here's an image if you still can't find it:

Don't set it to "J" like it's shown in the image, set it to "G" which is what you are sending to the game clients.

Do you have a mapped key, where the hotkey is G but the output to the game is N?

If you do, and Keymaps are enabled, then you will push G and the game will receive N.

I had a similar problem a while ago only on one set of toons and it was that the "Load out of date addons" wasnt selected for those toons and isboxer was out of date.

Finally this makes sense. The Key Combination for Click-to-Move ON set in Game Helpers is Shift+Alt+N. So, with the ISBoxer addon disabled when you press the Interact with Target's Hotkey, it sends Shift+Alt+N (among other things) to WoW (expecting the addon to interpret it) to turn Click to Move ON. Because of the way WoW handles keystrokes with modifiers, if nothing is bound to Shift+Alt+N it gets interpreted as unmodified N.

Suggestion to OP: I would have been helpful if you had also mentioned that neither inviting, follow or assist worked either.
