Open up the Toolkit, and in the upper left pane expand Character Sets, then click on your Character Set.

You'll notice in the bottom pane, under the "General" tab (which is the default view), there are options which you can check.
The third one down is for the "In-game ISBoxer GUI toggle hotkey".

I have mine set to: Ctrl + Alt + / because that is what Zanthor originally suggested in his "From Zero to Hero" thread.
But any unique combination will work.

I like to have a variety of Action Target Groups.

Usually there will be one for the entire team.
There will be another for just the Death Knights.
With only the one healer, I wouldn't make one for the Paladin... but if the composition was 3x DK's + 2x Healers, then a Healer ATG would be useful.

When you're in game, press your GUI toggle hotkey.

I believe Repeater Regions are the second tab.
There is a window for "Target", with an arrow to the right of it.
Clicking this arrow brings up valid targets... which can be Slot Number or a pre-defined ATG.

The healing Repeater Region, is set to Slot 3, because that is my Paladin's slot.
The mouse-over (clickable) stuff for the DK's can be set to their ATG.

Once the target is set, you have the option of KVM Mode (Block Local Click).
This can be good, if you only want the target (set, as per above) to receive the click.
I don't really want my DK's to drop their target, when the Grid frame is clicked upon... so I use the block local click option for that Repeater Region.
Even if I'm on the Paladin's screen (with Block Local Click enabled), because the Paladin is the target, she will get the clicks sent to her.
If you want KVM (Block Local Click), set that first then choose the target, because the target is reset to nothing when you check that box.

Next you click the button to create the repeater region.
Drag it over whatever you want.
Noting that the shaded red area is what will be "repeated", not the border of the Region.

Once it is in place, click on the sync button... which will place the region in the exact same place on every screen.
Then click "Save as Auto" or whatever the default option is.
The default save, will auto-load when the team is loaded.
If you change the save name, then it is not auto loaded, and must be manually started each time.

I have a variety of Repeater Regions.
There is one over the Jamba Item Bar (mine is five buttons across and two buttons tall).
I have Healthstones in the same position on every screen, with a Repeater Region over those.
Same thing for the Summoned Mage Cake; which is also hotkeyed.

There are four buttons for the DK's.
The Pally has identical graphics, but a blank macro in those places: Raise Dead, Anti-Magic Shell, Icebound Fortitude, Pillar of Frost.
The Raise Dead can only be clicked, there is no hotkey.
The other three have a hotkey or can be clicked because of the repeater region.
No real reason for not having a hotkey on Raise Dead; there was on originally but I took it out for some reason that eludes me now...

Basically put a Repeater Region over anything that you might want to click on the screen.
Where a click is good enough...

If you want targeting, in addition to the click...
You'll be better off with a Click Bar, which is essentially a click activated mapped key.
