If you want any of the advanced things, you pretty much need HKN (free) or IS Boxer (subscription). Keyclone (one-time license per computer that lasts forever) has all of the basics but is missing a lot of the advanced stuff.

IS Boxer and HKN will let you mouse broadcast, which is important if you want to aim Hurricane/Force of Nature/Blizzard/Flame Strike/Death & Decay etc. The mouse broadcasting is good for selecting talents or clicking portals.

IS Boxer has a small learning curve. Once you understand how the pieces work, it's pretty easy to figure out (or ask and interpret someone's instructions) on how to accomplish something. The wizard gets you automatic FTL assist, which is the best assist method.

All three products have excellent support, both from the users here and the authors. KC has a 24 hour trial, IS Boxer has a 7 day trial and HKN is free.

If you don't plan on doing the mouse broadcasting Keyclone has all of the basics. You'll probably go with a Focus or Target based assist, which is workable. You can set up an FTL assist with KC, but its a bit confusing. You can round-robin, blacklist, whitelist keys etc. It has most of the features that you would want.

If you want a bit more, HKN is free if you're comfortable with scripts (altering the example script or eventually writing your own), or IS Boxer has the toolkit with a wizard and lots of examples.
