If you want it to use an action button on slot 1, you need to make the action button in the game (ISBoxer will not do this for you, but will do all the hidden macros you want), and use a Keystroke Action as opposed to a WoW Macro Action. You can press 1 and have it do a WoW Macro Action that presses something else, like Q W E R T Y, [ ] \ and so on. I will add support for this kind of feature soon. This is usually the source of confusion -- thinking that the button you push is the button that needs to be assigned to the WoW Macro Action.

It is much easier to help you with your configuration if you share your config. Referring back to our conversation the other day in the chat channel, I think your Mapped Key probably would have worked better if it used only one key instead of two, and if you were using WoW Macro Actions assigned to 1,2,3, etc then see the first paragraph -- and we could have tested the macro in the game without the hotkey potentially interfering.
