You can definitely get multiple instances of LotRO to run on one PC. I have only ever tried 2 at a time though. If you have made a copy of your Lord of the Rings online folder inside of the Turbine folder and renamed it(example: LotRO 2ndClient), made a copy of the LOTROinvoker.exe file inside of your renamed LOTRO folder, rename it (example: Client 2) and then dragged that to desktop you should be able to run 2 instances now, make sure you have both of the login windows opened and ready to login. If you do one and enter the world before you have the second one started and ready for login, you will usually get the message "LotRO is already running, would you like to bring it to the front?" message. Also I am running a Core2Duo e6750 over clocked to 3.66Ghz with 8G of ram with the paging file size minimized and then disabled. I also set the CPU affinity of each LotRO client so each one is on a separate core independent of each other. This has reduced the overall % of CPU usage during play compared to having both clients utilizing both cores as the system sees fit. The one issue I do have is that I am using Keyclone to broadcast my keys to my third character on a seperate PC and would really like to get my second Instanced character more involved. Is there a utility I can use to lock that window in the forefront so that it accepts the broadcast from Keyclone??
Okay, quick question. Do you have totally different accounts? By this, I mean do you have 2 different user names to login with? I've tried this method in other variations, such as copying the folder to another drive, to the desktop and directly to c:. No good. I have 1 Turbine user name with 2 accounts attached to it. I'm wondering if that is the problem.

Elistrea does have multiple account logins.

Yes you will need a separate account for each instance of the game you wish to open.

I have a turbine login name, lets call it: mylogin and under this turbine login, I have 2 accounts, account01 and account 02. Does this work or do I need to setup mylogin1 and mylogin2 on the turbine site?

Not sure I follow exactly what you mean, but I have 2 copies of LotRO with two independent product keys, in order to log into two instances at the same time.

Default Elistrea's Method is a CONFIRMED work!

I just copied my main lotro directory and it does in fact allow me to
run two instances of lotro on one computer. Here are some basic step by
    step instructions:

    1. Go into my computer and find where you installed your Turbine game folder

    2. Open said folder and right click on the lord of the rings online folder (select copy and paste within your turbine folder)

    3. Depending on the machine, you may have to wait a bit for it to copy.

    4. When it is finished, open the new folder and find the Turbine
Launcher icon (should be about 2.11 mb) and copy that to the desk top.

    5. Click your original copy of lotro to start the program, then click
the new turbine launcher (your computer may ask you to unblock or block
turbine so please allow it to access the internet.

    6. You should be able to play two instances of lotro on one box now. I
am now dual boxing my warden and using a lore master for help. Cheers

Is the macro system still limited to /follow ? Any target forwarding in the game ?

I'd really appreciate hearing about any macros people are using or how they are getting around not using them. I'm using Octopus across multiple PCs and am looking at running 2 characters on separate PCs.
