I am new to this whole multiboxer. My secondary team of 3 druids is getting close to level 20 at which point I want to run them all as cats.
What do I need to do to get the two following druids to move within melee attack range?

Have you tried using interact with target? Not sure if it's the best method but when I used melee I'd just click that for my guys to run in melee range and start autoattacking. Only annoying thing is if the target moves you need to press it again and occasionally one of my guys would run off in a random direction.
I'm not very experienced with melee so there might be a better method.

It is a macro? Can I do all 3 steps in the same macro or will isboxer take care of the first step for me, and all I need is the second set of macros?
/assist character 1
/interact
/startattack
?
/interact
/startattack

Your interact with target command is found in ISBoxer under Keymaps -> Base Hotkeys. Set that to a particular bind both on ISBoxer and on all 3 of your in-game wow keybinds and it should work.
You can also modify your keybinds in ISBoxer to automatically IWT on individual keys, so it would essentially bypass the need for this individual key but these keybinds should not be shared with ranged abilities (spells outside of cat/bear form).
