Two possibilities I can think of.
If if works, when you're doing your DPS spam but not when you manually press it...
Then your manual press mapped key, might be outputting a different keybind to the game than the working keybind which is sent by the DPS spam.
The other thing I'd check: Do you have Click to Move enabled or not?
Basically, whether you send IWT successfully or not, if CTM is disabled your characters cannot move in response to the IWT command.
Your DPS spam might enable CTM for the slave (or maybe all melee) characters, but by default you have CTM set to off.
IS Boxer does have two IWT type commands, for manual use.
Both generated by the wizard.
One of these enables CTM, sends IWT, and disables CTM.
The other just sends IWT, without adjusting your CTM.
Look at your assorted keymaps/mapped keys that do the IWT action.
Check that CTM is not being toggled off.
You can toggle it on via: /console AutoInteract 1
And toggle it off via: /console AutoInteract 0
You can also manually set it, via clicking/unclicking a box... in Escape > Interface Options > Mouse Options

While the video still contains useful information, the process has changed slightly and the video hasn't been updated to reflect those changes. I suggest you just follow the written directions on the wiki page:
http://isboxer.com/wiki/WoW:Interact_with_Target
It literally comes down to:
Setting a key binding in game
Setting a hotkey for the IWT Key Map in ISBoxer (doesn't have to be the same key you just used, but it can be)
Setting a key for the IWT Variable Keystroke in ISBoxer (the same key you used in game)
Exporting your settings

its still not working, are you useing repeater to bring up the quest menus on all your chars? is there a way you can use teamviewer to help me? i uninstalled it and reinstalled it following those directions but still the same

No, I probably use my IWT every time I speak to NPCs when multiboxing and, for the most part, even when I'm playing solo.

No.

Reinstalling ISBoxer won't fix anything.
Step 2 tells you to turn your Key Maps off. Did you turn your Key Maps back on when you were finished?
If so, what happens when you turn your Key Maps off and press your IWT in-game key binding?

To me, this sounds like you've incorrectly set the IWT key binding on that character. If you visually check that character's key binding and that isn't the issue, then the only way to move forward with this is for you to share your configuration (http://isboxer.com/forum/viewtopic.php?f=14&t=13).

Looking at your profile, you've modified the default IWT Mapped Key and hardcoded it to send G to only your active client (Window:Current). There are numerous ways to fix this, but the quickest way would be to change the target from Window:Current to Window:All w/ Current.

Assuming you've exported your settings to Inner Space and your Key Maps are enabled (SHIFT+ALT+M), when you press ALT+G ISBoxer is sending G to all of your clients. If G is not set as the key binding in World of Warcraft to perform Interact with Target (in each and every game client) then nothing will happen.

That's because the default in-game key binding for your talent pane is N. You seem to not be understanding that you need to assign an Interact with Target key binding in World of Warcraft because this action is not bound to anything by default in the game itself. Here are the instructions you should have first followed direct from the Wiki page (this is also shown in the video you watched):
Finding Interact with Target in the in-game Key Bindings window
Enter the game
Turn Key Maps off (Shift+Alt+M)
Turn Repeater on (Shift+Alt+R)
Open up the in-game menu with Video, Interface, Key Bindings and other options (Escape)
Click on Key Bindings
Locate the Interact with Target key binding, under Targeting
Set the Interact with Target key binding to your desired key combination, such as 'G'
Turn Repeater back off (Shift+Alt+R)
Turn Key Maps back on (Shift+Alt+M)
Here's an image if you still can't find it:

Don't set it to "J" like it's shown in the image, set it to "G" which is what you are sending to the game clients.

Do you have a mapped key, where the hotkey is G but the output to the game is N?
If you do, and Keymaps are enabled, then you will push G and the game will receive N.

I had a similar problem a while ago only on one set of toons and it was that the "Load out of date addons" wasnt selected for those toons and isboxer was out of date.
Finally this makes sense. The Key Combination for Click-to-Move ON set in Game Helpers is Shift+Alt+N. So, with the ISBoxer addon disabled when you press the Interact with Target's Hotkey, it sends Shift+Alt+N (among other things) to WoW (expecting the addon to interpret it) to turn Click to Move ON. Because of the way WoW handles keystrokes with modifiers, if nothing is bound to Shift+Alt+N it gets interpreted as unmodified N.
Suggestion to OP: I would have been helpful if you had also mentioned that neither inviting, follow or assist worked either.
